The ubiquitin-proteasome system (UPS) removes normal but no longer needed proteins as well as abnormal proteins in the cell. It is generally believed that the 26S proteasome (i.e. 19S proteasome associated 20S proteasomes) is in charge of the degradation of targeted house-keeping proteins. UPS functional insufficiency has been implicated in the genesis of congestive heart failure. Measures to enhance proteasome proteolytic function are currently lacking but would both be extremely beneficial to decipher the pathogenic role of proteasome dysfunction and have great therapeutic potentials. The 11S proteasome can be formed by hetero-polymerization of PA28alpha and PA28beta. The association of 11S proteasomes with the 20S proteasome is believed to play an important role in antigen processing. The present study revealed that in cultured neonatal rat ventricular myocytes PA28alpha overexpression (PA28alphaOE) stabilized PA28beta, increased 11S proteasomes, and caused significant decreases in the protein level of a previously validated proteasome surrogate substrate, a modified green fluorescence protein (GFPu). Pulse chasing revealed a proteasome-dependent destabilization of GFPu protein by PA28alphaOE. To verify these findings in intact animals and investigate the impact of PA28alphaOE on the heart, stable transgenic mouse lines were created to allow doxycycline-suppressible cardiomyocyte-restricted (CR-) PA28alphaOE. CR-PA28alphaOE increased PA28beta and 11S proteasomes in the heart but various analyses failed to detect cardiac abnormalities, at least during the first year of age. Cross-breeding CR-PA28alphaOE mice with a proteasome functional reporter (GFPdgn) mouse revealed that GFPdgn protein levels were significantly decreased by CR-PA28alphaOE via a post-transcriptional mechanism. Furthermore, suppressing CR-PA28alphaOE by doxycycline markedly attenuated the decrease of GFPdgn protein levels in the heart; correspondingly, CRPA28alphaOE induction by doxycycline
